Corticosterone exerts its neuroprotective effects through inhibiting the glutamate uptake, promoting the glutamate-induced microglia activation. The purpose of this study is to determine whether the low efficacy of corticosterone in improving PD is related to microglia activation. Mice were given with α-Mangostin, which could prevent microglia from activating. We found that α-Mangostin treatment could enhance the efficacy of corticosterone in preventing PD. In the corticosterone-treated PD mice, the expressions of inflammation cytokines and microglia activation markers were decreased and glutamate-induced microglia activation was also decreased. α-Mangostin treatment could prevent microglia from activating, and glutamate-induced microglia activation.
